Unlock Characters
Baki - Complete Gaara, Temari, and Kankuro's first and second missions.
Bando - Complete Kabuto's first and second missions.
Itachi - Complete Sasuke's first and second missions.
Kabuto - Complete Baki.
Kagura - Complete Bando's first and second missions.
Kurenai - Complete Kiba, Hinata, and Shino's first and second missions.
Kisame - Complete Itachi, Guy, and Asuma's first and second missions.
Komachi - Complete Story Mode.
Anko - Complete Orochimaru's first and second missions.
Asuma - Complete Ino, Shikamaru, Choji, and Kurenai's first and second missions.
Orochimaru - Complete both Jiraiya and Tsunade's first and second missions.
Towa - Complete Story Mode.
Second State Sasuke - Beat the first and second missions then the last three missions with Jonin rank or higher.
Awakened Hinata - Beat the first and second missions then the last three missions with Jonin rank or higher.
Ultimate Nine Tailed Naruto - Beat the first and second missions then the last three missions with Jonin rank or higher.

Unlock Modes
Kumite Mode - Come in first on score attack mode.
Survival Mode - Come in first place on time attack mode.
2P Survival - Get al least Jonin in 2P Time Attack.
2P Time Attack - Get at least Jonin in 2P Score Attack.

Story Viewer
This is the last bar in Extras. It allows you to view story mode cutscenes without doing the story mode:

Story Viewer - complete story mode on level 4 difficulty.

Unlock All Characters Through Missions
Remember that you must finish the Story Mode on Difficulty 2 or higher before attempting to unlock characters through missions. Just change the game settings to Difficulty 2 and it affects all modes:

Anko - 15% mission completion for Orochimaru.
Asuma - 30% combined mission completion for Ino, Shikamaru, and Choji.
Baki - 30% combined mission completion for Gara, Temari, and Kankuro.
Bando - 20% mission completion for Kabuto.
Itachi - 30% combined mission completion for Kakashi, Asuma, and Kurenai.
Kabuto - Unlock all characters except for UNT Naruto, SS Sasuke, A Hinata, Bando, and Kagura.
Kagura - 30% combined mission completion for Kabuto, Bando, and Tsunade.
Kisame - 30% combined mission completion for Guy, Asuma, and Kurenai.
Komachi - Finish the Story Mode on Difficulty 2 or higher.
Kurenai - 30% combined mission completion for Hinata, Kiba, and Shino.
Orochimaru - 20% combined mission completion for Jiraiya and Tsunande.
Towa - Finish the Story Mode on Difficulty 2 or higher.
Yugao - 30% combined mission completion for Baki, Komachi, and Towa.

Unlock Voices
You can unlock a characers voice by completing: The first rival match Both of the 2 vs 2 matches and The 1 vs 3 match:

Character's Voice - Complete the 1st two missions, the 1 vs 3, and the Second 2 vs 2.

Unlock Stages
Final Valley - Beat U.N.T Naruto and Sasuke SS Last Missions (difficulty 2).
