Enter using D-Pad while in the Helicarrier between levels:

[Right], [Right], [Right], [Up], [Down], [Left] - Unlock Sandman
[Left], [Left], [Right], [Up], [Down], [Down] - Unlock Venom
Hints
Unlock Hero Concept
Finish the game and then finish Tokyo area 1.

Unlock Carnage
Finish the Co-Op mode.

Unlock Sidekicks
Either get to a certain point in the game or buy them from the shop:

Black Cat - Beat the tutorial level.
Doctor Octopus - Buy for 500 tokens in the shop.
Green Goblin - Buy for 750 tokens in the shop.
Blade - Buy for 1000 tokens in the shop.
Sandman - Buy for 1250 tokens in the shop.
Venom - Buy for 1500 tokens in the shop.
